CellTram® 4r Air/Oil

The CellTram 4r Air and CellTram 4r Oil are manual microinjectors suitable for a broad range of life science applications. The CellTram 4r Air is a pneumatic injector for oil-free micromanipulation and gentle holding of cells in suspension. Due to its excellent performance, it is also highly suitable for uptake and injection of cells (e.g., of sperm or ES cells). For this application, its performance is comparable to that of oil-filled injectors. The CellTram 4r Oil is a hydraulic, oil-filled microinjector that provides more direct sample control and can generate higher pressures than an air system. It is optimized for leakage-free work and easy oil filling. Both models can be used with all common micromanipulation systems yet they develop their full potential with Eppendorf manipulators. The CellTram 4r Air/Oil injectors are for research use only.

Optimal protection for your cells
Minimizing mechanical trauma to your sample is critical for achieving optimal results. The new capillary holder 4 (slim shape) in combination with Eppendorf manipulators allows for injecting with straight capillaries at angles lower than 15°. This helps to achieve highest survival rates for your developing embryos.

Frequently Asked Questions

I read in the manual that the piston position of the CellTram 4 Air has an influence on the responsiveness in sample control. What are the recommended working positions / piston settings with the CellTram 4 Air?‌

The lower the selected piston position is, the more direct the responsiveness of your sample in the capillary is. This means, at piston positions of 1-3 the rotation of the drive wheel causes a faster or stronger movement of the sample within the capillary. In case you choose a piston position of 7-9, the sample in the capillary is moved less or more slowly per rotation of the drive wheel. Since many users have individual preferences in their injector responsiveness, we recommend to identify your optimal piston position for your particular application (e.g., holding of oocyte, transfer of sperms or cells, etc.) by testing different piston positions.

My CellTram 4 Air does not react as expected when turning the fine or coarse drive wheel. What could be the reason?‌

Please check if any fluid/moisture is inside the injection tube or capillary holder. If yes, remove the fluid by flushing air several times through the injection tube or capillary holder, e.g. with the filling- and cleaning set (Order no. 5196 043.006), and let dry overnight. Please check if all connections (e.g., at the grip head, at the capillary holder, at the instrument itself) are screwed on tightly. Also check the o-rings inside the grip head and exchange them in case of any scratches. Make sure that the outer diameter of your capillary tubing fits to the size of the grip head. Our standard grip head of size 0 is designed for capillaries of 1-1.1 mm in outer diameter.

When I use the ventilation valve of the CellTram 4 Air to obtain equilibration, my cells within the capillaries get strongly sucked deeper into the capillary. Is the ventilation valve not working well?‌

The ventilation valve allows for pressure equilibration of the system, meaning the instrument cylinder and injection tube, and ambient pressure. Beside the pressure equilibration in the system, the physical capillary forces may result in movement of liquid. We therefore recommend not performing the equilibration when cells are in or directly in front of the capillary.

Can I convert the CellTram 4 Air to a hydraulic (oil-filled) injector?‌

No, the CellTram 4 Air should be not filled with oil or any other fluid.

In your catalog you recommend the pneumatic injector CellTram 4 Air for the holding of oocytes and early embryos. Can you also hold these samples with the oil-filled injector CellTram 4 Oil?

As the devices differ in their maximum pressure, we in general recommend the CellTram 4 Air (max. pressure 3,000 hPa) for gentle holding of oocytes.
For special applications (e.g., biopsies) where a stronger vacuum may be required for reliable attachment of the sample, using the CellTram 4 Oil (max. pressure 20,000 hPa) for holding is an alternative.

My CellTram 4 Oil does not react immediately after turning the dial. Why?‌

Please check the system for any air bubble. If you detect any little air bubble within the cylinder, tubing or capillary holder, remove the air by refilling the system with oil using the filling valve. The grip head should be loosened a bit when re-filling with oil. Inserting the glass capillary into the grip head make sure that you produce no air bubble. After inserting the capillary turn the grip head to fix well.

Can you use different types of oil for filling the CellTram 4 Oil on the one hand and for covering the cells on the other hand?‌

Yes, this is possible, as both liquids usually do not come into direct contact.

Is it necessary to use paraffin oil for the CellTram 4 Oil, or is it also possible to use silicone oil?‌

You can use paraffin or silicone oil, depending upon the viscosity required for your application. Please clean the device very carefully when changing the oil.

We recommend the following light oil from Sigma. Product-No. M-8410.

Can I use the injection tube Air with the CellTram 4 Oil (and vice versa)? What effects might I see? How do the injection tubes distinguish?‌

The injection tube Air has a smaller inner diameter than the injection tube Oil. To guarantee the requested performance of the injectors, please use the two different injection tubes only in combination with their appropriate injector. The injection tube Oil is labeled with a blue ring mark at the connector pieces whereas the injection tube Air is labeled with white ring marks.

Can I extend the injection tube (1.3 m) of my CellTram 4r?

Yes, two injection tubes can be connected with the help of a tube coupling. Please note that you also have to order an additional injection tube (Injection tube Air: 5196 021.002; Injection tube Oil: 5196 042.000) in addition to the tube coupling (order number: 5176 220.009).

Can I order the injection tube of the CellTram 4 Oil/Air, which is connected with the capillary holder 4 (5196 041.003) separately?

The injection tubes for CellTram 4 Air and CellTram 4 Oil can be ordered separately.
Please note, that due to differences in the inner diameter the injection tubes should be only used for the correspondent injector as indicated. The order numbers are:
Injection tube Air (ID: 0.5 mm, white ring mark): 5196 021.002; Injection tube Oil (ID: 1 mm, blue ring mark): 5196 042.000.

How do you remove pieces of broken glass capillaries from the capillary holder?

For this purpose we included a cleaning stylet with the capillary holder. In case it is lost, we offer a kit, the so-called O-ring-set 4 for grip head sets 4 (Order no. 5196 086.007) that contains this tool. Alternatively, you can also use a thin metal wire.

What is the outer diameter of the capillary holder 4?

The outer diameter of the capillary holder 4 is 4 mm, as it is of our predecessor, the so-called 'universal capillary holder'.

Which grip head is included in the delivery package of the CellTram 4 Air/Oil?‌

Grip head size 0. It can be used for capillaries with an outer diameter of 1,0-1,1 mm. 

How can the different grip head sizes be distinguished?‌

The grip heads have different numbers of notches. The grip head 4, size 0 has no notch at all; the grip head 4, size 3 has, for example, three notches.

Where can I find the CE-certificate of the CellTram 4r Air/Oil?‌

A CE-certificate for CellTram 4r (for research only) is not available, as no certification directives for manual injectors in research exist.

What does the Essential Check of a microinjector/micromanipulator include?‌

Basic cleaning and checks, oil change, analysis of error statistics and a test run of the system at the end.

What does Advanced Maintenance of a microinjector/micromanipulator include?‌

A comprehensive service including basic cleaning of the outside and the interior assemblies, checking keyboard and display, a leakage test, oil change, software upgrade, motor module test, analysis of error statistics and a test run of the system at the end.
